چکیده

39 The nucleus accumbens (NAc) has been shown to play a key role in the brain’s response to methylphenidate 40 (MPD). The present study focuses on neuronal recording from this structure. The study postulates that repetitive 41 exposure to the same dose of MPD will elicit in some rats behavioral sensitization and in others tolerance. 42 Furthermore, the study postulates that NAc neuronal activity recorded from animals expressing behavioral 43 tolerance after repetitive MPD exposure will be significantly different from NAc neuronal activity recorded from 44 animals expressing behavioral sensitization after repetitive MPD exposure at doses of 0.6, 2.5, 5.0, and 10.0 mg/kg. 45 To test this, behavioral and neuronal activity was recorded concomitantly from the NAc of freely behaving 46 adolescent rats (P-40) before and after acute and repetitive administration of four different MPD doses. Comparing 47 the acute MPD effect to the repetitive MPD effect revealed that the acute response to MPD exhibited dose response 48 characteristics: an increase in behavioral activity correlated with increasing MPD doses. On the other hand, 49 following repetitive MPD exposure, some animals exhibited attenuated behavior (tolerance), while others 50 exhibited further increases in the recorded behavior (sensitization). Moreover, the neuronal activity following 51 repetitive MPD exposure recorded in animals exhibiting behavioral sensitization was significantly different from 52 neuronal activity recorded in animals exhibiting behavioral tolerance. This implies that when studying the effects 53 of repetitive MPD administration on adolescent rats, it is advisable to simultaneously record both neuronal and 54 behavioral activity, and to evaluate all data based on the animals' behavioral response to the repetitive MPD 55 exposure. 56 57

Behavioral and neurochemical effects of 5-(4-[4-(5-Cyano-3-indolyl)-butyl)-butyl]-1-piperazinyl)-benzofuran-2-carboxamide (EMD 68843): a combined selective inhibitor of serotonin reuptake and 5-hydroxytryptamine(1A) receptor partial agonist.

5-(4-[4-(5-Cyano-3-indolyl)-butyl)-butyl]-1-piperazinyl)-benzofuran-2-carboxamide (EMD 68843; vilazodone) is a novel compound with combined high affinity and selectivity for the 5-hydroxytryptamine (5-HT) transporter and 5-HT(1A) receptors. EMD 68843 was tested as a prototype compound, which benefits from dual pharmacological effects that could increase extracellular 5-HT to levels higher than ...

Effect of acute and chronic treatment with QCF-3 (4-benzylpiperazin-1-yl) (quinoxalin-2-yl) methanone, a novel 5-HT(3) receptor antagonist, in animal models of depression.

The serotonin type 3 (5-HT(3)) receptor is unique among the seven recognized serotonin receptor "families". The existence serotonin type 3 receptor (5-HT(3)) in neuro-anatomical regions stimulated the research interest for novel therapeutic targets such as anxiety, depression, nociception and cognitive function. In the current study, (4-benzylpiperazin-1-yl) (quinoxalin-2-yl) methanone (QCF-3),...

Inhibition of monoamine oxidases desensitizes 5-HT1A autoreceptors and allows nicotine to induce a neurochemical and behavioral sensitization.

Although nicotine is generally considered to be the main compound responsible for addictive properties of tobacco, experimental data indicate that nicotine does not exhibit all the characteristics of other substances of abuse. We recently showed that a pretreatment with mixed irreversible monoamine oxidases inhibitors (MAOIs), such as tranylcypromine, triggers a locomotor response to nicotine i...

Antidepressant Potential of 5-HT3 Receptor Antagonist, N-n- propyl-3-ethoxyquinoxaline-2-carboxamide (6n)

The present study was designed to evaluate the antidepressant potential of 5-HT3 receptor antagonist N-n-propyl-3-ethoxyquinoxaline-2-carboxamide (6n). The compound '6n' with optimum log P and pA 2 value identified from a series of compounds synthesized in our laboratory was subjected to forced Swim Test (FST) (1, 2, and 4 mg/kg, i.p) and Tail Suspension Test (TST) (1, 2, and 4 mg/kg, i.p.). Th...

Age-dependent biphasic changes in ischemic sensitivity in the striatum of Huntington's disease R6/2 transgenic mice.

We used the oxygen/glucose deprivation (OGD) model of ischemia in corticostriatal brain slices to test the hypothesis that metabolic deficiencies in R6/2 transgenic Huntington's disease (HD) mice will impair their recovery from an ischemic challenge. Corticostriatal extracellular field excitatory postsynaptic potentials (fEPSPs) were evoked in transgenic and wild-type (WT) mice in three age gro...
